Personalized neoantigen target discovery - ImmunoEngine for personalized immunotherapies

The myNEO workflow discovers and prioritizes neoantigens for individual patients using our ImmunoEngine pipeline. The workflow is IVD-ready. Starting from tumor and matched healthy sequencing data, it analyzes variant expression and translation, predicts MHC class I and II presentation, scores CD4+ and CD8+ immunogenicity, and selects a prioritized neoantigen set. Results are reviewed in neoHUB. Selected neoantigens can proceed to myCONSTRUCT and myRNA for product design.

What myNEO delivers

The personalized workflow has a turnaround of five business days, from receipt of sequencing data to the prioritized neoantigen selection. Optional myCONSTRUCT and myRNA design can follow within one day:

  • Required inputs: whole-genome and RNA sequencing of the tumor, and whole-genome sequencing of a matched healthy sample, usually blood
  • Variant expression analysis
  • Translation analysis
  • Presentation prediction: identification of MHC-presented epitopes on both MHC class I and MHC class II molecules
  • Immunogenicity prediction: identification of presented epitopes recognized by both CD4 and CD8 immune cells
  • Final epitope selection: based on antigen score, clonality, functional relevance, and immune-escape likelihood
  • Data visualization: web-based reporting in neoHUB for navigating complex datasets
  • myCONSTRUCT: next-generation string-of-beads design (optional)
